摘要
目的探讨IL-1基因IL-1B-31/-511位点的多态性与胃癌发病风险、IL-1β分泌水平的关系。方法使用基因芯片检测技术,对胃癌手术患者及消化科十二指肠球部溃疡患者全血标本结合PCR体外扩增方法,检测人IL-1B基因-31和-511位点的基因多态性(IL-1B-31C/T和IL-1B511C/T)。比较IL-1B基因-31和-511位基因多态性与各种胃癌临床病理参数的关系。结果IL-1B-31及IL-1B-511位点的T携带子在胃癌组与十二指肠球部溃疡患者比较,分别增高胃癌发病风险OR=3.675(95%CI:1.359-9.940)、OR=3.111(95%CI:1.163~8.322)。不论在胃癌组还是在十二指肠球都溃疡组IL-1B-31T携带子、-511T携带子基因型患者血清IL-1I]水平均高于IL-1B-3C/C、-511C/C基因型患者,且IL-1B-31T携带子、-511T携带子基因型在胃癌组患者的IL-1β水平较十二指肠球部溃疡组患者高。结论IL-1B-31T基因型和IL-1B-511T基因型增加胃癌发病风险,且与IL-1β分泌水平有关。
Objective To investigate the correlation between polymorphism of interleukin-lB genes and risk of gastric cancer, the serum level of IL-β. Methods Crene analysis and biochip technique was used to analyze the - 31 and - 511C/T polymorphisms of the IL-1B DNA from the peripheral blood of 58 patients with gastric carcinoma, 24 patients with duodenal ulcer from the first affiliated hospital of Suzhou university. Results IL-1B-31, - 511 polymorphism (carriers of IL-1B- 31T, -511T) was correlated with the increased risk of primary gastric cancer[carriers of IL-1B-31T OR=3.675(95%CI1.359-9.940) (X^2=6.917,P=0.027)] [carriers of IL-1B-511T OR=3.111 (95 % CI1. 163 - 8. 322) ( X^2 = 5.316, P = 0. 012) ]. The serum level of IL-1β increased in carriers of IL-1B-31T,IL-1B-511T in patients with gastric carcinoma or duodenal ulcer. Conclusion The IL- 1B-31T, - 511T genotypes increase the risk of gastric cancer and are associated with the production of IL-1β.
