胰岛素治疗对糖尿病大鼠牙槽骨中OPGmRNA水平的影响 被引量:1

摘要 目的:研究胰岛素治疗对糖尿病大鼠牙槽骨中骨保护素(OPG)mRNA水平的影响,探讨血糖水平的变化在牙槽骨改建中的意义。方法:将12只大鼠采用静脉注射链脲佐菌素建立糖尿病模型,随机分为治疗组和对照组。治疗组给予胰岛素皮下注射,对照组未作干预。8周后处死大鼠,提取其左下磨牙区牙槽骨组织总mRNA,应用RT-PCR检测OPGmRNA表达水平。结果:胰岛素治疗组较对照组血糖明显降低(P<0.05),治疗组的OPGmRNA表达水平高于对照组(P<0.01)。结论:胰岛素治疗在使糖尿病大鼠血糖降低的同时,可使牙槽骨OPGmRNA表达水平升高,提示良好地控制血糖可使糖尿病大鼠的牙槽骨吸收减慢。 Objective: To investigate the effect of insulin therapy on the mRNA expression of osteoprotegerin (OPG) in the alveolar bone of diabetes meUitus rats, and assess the meaning of the change of blood glucose level in alveolar bone resorption. Methods: Twelve SD rats were injected intravenously with streptozotocin (STZ) to induce diabetes mellitus. These rats were divided into 2 groups randomly, of which one was as treatment group and the other was as control group. The rats of treatment group were injected hypodermically with insulin. The rats of control group were not provided intervention. All rats were executed after 8 weeks. The total RNA in the alveolar bone of molar region of the left jaw was extracted. Detect the expression of OPG mRNA with RT-PCR. Results: The blood glucose level of treatment group was lower than that of control group(P〈0.05). The expression of OPG mRNA in alveolar bone of treatment group was higher than that of control group (P〈0.01). Conclusion: The treatment of insulin can reduce the blood glucose of diabetes meUitus rats, and it also can increase the mRNA expression of OPG in the alveolar bone. This result suggests that reducing blood glucose may slow the litus rats. ve speed of alveolar bone of diabetes mellitus rats.
