新生鼠高浓度氧肺损伤临界值的初步研究 被引量:1

摘要 目的探讨持续吸入常压下不同浓度氧引起新生鼠肺损伤的临界时间。方法4组新生鼠分别暴露于>95%O2、60%O2、40%O2和正常空气(21%O2)中,取暴露12、24、48和72h的肺组织,在光镜下和透射电镜下观察其病理改变。结果新生鼠吸入>95%O212h、60%O248h或40%O272h后,均出现肺组织的病理改变,光镜下可见微血管扩张、充血,肺间隔增宽,肺泡内有蛋白渗出,炎症细胞浸润,出血,肺组织结构紊乱,肺大泡数量增多等;透射电镜下可见内皮细胞线粒体肿胀,内质网肿胀、破坏,吞饮小泡增多,Ⅱ型肺泡上皮细胞脱落等;随着吸氧浓度的增高和时间的延长,肺损伤程度加重。结论吸入95%以上O212h,60%O248h,或40%O272h均可能引起新生鼠肺损伤,适当控制吸氧浓度和时间有助于减少肺损伤的发生,减轻肺损伤的程度。 Objective To study the effects of the oxygen concentration and time of hyperoxic exposure on lung in neonatal rats. Methods The neonatal rats who were less than 12 hours old were exposed to 〉95%O2, 60%O2, 40%O and normal air (21%O2) respectively in 4 group. The rats lungs tissues were obtained in all the groups 12, 24, 48 and 72 hours after the exposure to the different concentration oxygen. The pathologic changes in the lung were observed under light microscope and transmission electron microscope. Results The pathologic changs occured in the lungs of the neonatal rats who were exposed to 95 %O2 for 12 hours, 60%O2 for 48 hours and 40%O2 for 72 hours. The pathologic changs included microvessel hyperemia, innammatory cell infiltration, hemorrhage, swollen endoplasmic reticulum and mitochondrion, and deciduous Ⅱ type alveolar epithelial cells . The lung injury aggravated with the increase in the concentration of inhaled oxygen and the inhalation time prolonging. Conclusion The exposure to 95%O2 for 12 hours, 60%O2 for 48 hours and 40%O2 for 72 hours can cause lung injury in neonatal rats. The proper control of the concentration of inhaled oxygen andd inhalation time is helpful to decreasing the incidence of lung injury and relieving lung injury.
