摘要
目的:探讨强骨宝方糖尿病模型鼠含药血清促进体外培养成骨细胞增殖的最佳时相与量效。方法:采用胰蛋白酶-Ⅱ型胶原酶消化法从1-2日龄SD大鼠颅盖骨中分离出成骨细胞,鉴定细胞后,用不灭活的不同时效(灌胃3d、5d,末次灌胃后1h、3h)、不同浓度(5%、10%、20%)的强骨宝方糖尿病模型鼠含药血清加入成骨细胞培养体系,培养48、72h,应用MTT比色法来检测其对成骨细胞增殖功能的影响。结果:各时相的强骨宝方糖尿病模型鼠含药血清对成骨细胞的增殖作用呈浓度依赖性,即20%>10%>5%;其中以3d末次灌胃后1h、20%的含药血清作用最明显,在成骨细胞培养48h、72h后均具有显著促进其增殖作用。结论:灌胃3d、末次灌胃后1h、20%不灭活的强骨宝方糖尿病模型鼠含药血清对成骨细胞增殖作用最明显。
Objective： To investigate the optimal time-phase and quantity-effect of Qianggubeo Formula contained serum of diabetes rat models in promoting the osteoplast multiplication in vitro culture. Methods： Adopting trypsinase-type Ⅱ collagen enzyme digestion, the osteoplasts were aparted from cranium of 12 day old SD rat, after cell identification, non-inactivated Qianggubao Formula contained serum of diabetes rat models in different time-effect （ intragastric administration 3d, 5d and lh and 3h after the last intragastric administration） and concentration （5%, 10% and 20% ） were added to osteoplast culture system. After cultured for 48h and 72h, the effects of Formula contained serums diabetes rat models on osteoplast multiplication funfioan were detected by MTT colorimetry. Results： The effect of Qianggubao Formula contained serum of diabetes rat models in different time-phase on osteoplast multiplication presented concertration dependent, I.e. 20% 〉 10% 〉 5% ; among the total, the effect of Qianggubao Formula contained serum was most obvious in the time of lh after the last time of 3d intragastric administration and with the concentration of 20%. After osteoplast cultured for 48h and 72h, they were presented significant promotion of multiplication action. Conclusion： The effect of non-inactivated Qianggubao Formula contained serum was most obvious in the time of 1h after the last time of 3d intragastric administration and with the concentration of 20%.
