摘要
[目的]体外研究不同剂量脂多糖(LPS,lipopolysaccharide)对过敏性哮喘患者细胞因子分泌的调节作用。[方法]随机选取过敏性哮喘患者8例,取其外周血3份,其中两份分别加入10μg、1μg的LPS进行24 h培养,设为高、低剂量实验组;另一份不加LPS同样培养,为对照组。ELISA法检测各组IL-4、IL-5、IL-10、IL-12及IFN-γ含量。[结果]3组间比较发现IL-4、IL-5、IL-10、IL-12生成量及IFN-γ/IL-4比值差异有统计学意义(P﹤0.05),而IFN-γ生成量差异无统计学意义。与其他两组相比,高剂量组的IL-4、IL-5的生成量显著降低(P﹤0.05),而IL-10、IL-12生成量及IFN-/γIL-4比值显著升高(P﹤0.05);低剂量组与对照组比较,IL-5显著性升高(P﹤0.05)。高剂量组IL-12与IL-4及IL-5呈负相关(r=-0.822,P﹤0.05;r=-0.768,P﹤0.05),与IL-10呈正相关(r=0.841,P﹤0.05);IL-10与IL-4及IL-5呈负相关(r=-0.812,P﹤0.05;r=-0.754,P﹤0.05)。[结论]LPS调节过敏性哮喘患者细胞因子分泌具有剂量依赖性,高剂量LPS能有效调节过敏性哮喘患者细胞因子的分泌,抑制Th2型优势免疫。
[Objective] To confirm the endotoxin dose-dependence of regulating the production of the cytokines of the peripheral blood in the allergic asthma patients. [Methods] The allergic asthma patients with the middle severity or above were selecteel at random and their heparinized venous peripheral blood were collected. The blood were cultured with the high (10 μg) or low ( 1 μg) dose of LPS for 24h, and the controle group was set up without LPS treatment. The cytokines were measured by the enzyme-linked immunosorbent assay. [ Results] (1) The differences of the levels of IL-4, IL-5, IL-10, IL-12 and IFN-γ/IL-4 among three groups were significant except IFN-γ. (2) The high dose group had higher levels of IL-10, IL-12, IFN-γ/IL--4 and lower levels of IL-4, IL-5 than the low dose group or control group (P〈 0.05) ; the level of IL-5 in the low dose group was higher than the control group (P〈 0.05), but the level of IFN-γ did not differ between them. (3) In high dose group, the concentration of IL-12 was negatively correlated to that of IL-4 and IL-5 (r = -0.822, P〈 0.05; r =-0.768, P〈 0.05), possitively correlated to that of IL-10 (r = 0.841, P〈 0.05) ; IL-10 was negatively correlated to IL-4 and IL-5 (r = - 0.812, P〈 0.05; r = -0.754, P〈 0.05) . [Conclusion] LPS can dosedependently modulate the cytokines secretion of the allergic asthma patients in vitro. High dose LPS lead to the increase in the levels of IL-10, IL-12 and the decrease in that of IL-4, IL-5, and inhibit the Th2 dominant immunity.
