摘要
目的探讨低氧对体外培养的人脐静脉内皮细胞胶原ⅩⅧ和内皮抑素表达的影响。方法取体外原代培养的人脐静脉内皮细胞,分为对照组、低氧组、复氧组,对照组细胞正常培养24h,低氧组细胞放入氧浓度为2.5%低氧箱中培养24h,复氧组细胞于低氧培养24h后继续正常培养24h,采用ELISA法和RT-PCR技术检测内皮细胞胶原ⅩⅧmRNA和内皮抑素的表达。结果各组内皮细胞中均存在胶原ⅩⅧmRNA和内皮抑素的表达。与对照组比较,低氧组细胞胶原ⅩⅧmRNA和内皮抑素的表达明显增加,复氧组细胞胶原ⅩⅧmRNA和内皮抑素的表达较低氧组细胞明显降低,但高于对照组。结论低氧增加人脐静脉内皮细胞胶原ⅩⅧ和内皮抑素的表达,低氧可能是影响胶原ⅩⅧ和内皮抑素表达的因素之一。
Purpose To investigate the effects of hypoxia on the expression of collagen ⅩⅧ and endostatin in human umbilical vein endothelial cells(HUVECs). Methods HUVECs primary cultured were randomly divided into control, hypoxic and reoxygenation groups and separately cultured in normoxic and hypoxic condition in vitro. The levels of endostatin in the culture media of HUVECs were measured by ELISA. The collagen ⅩⅧ mRNA expression in HUVECs was measured by RT-PCR. Results HUVECs could liberated endostatin into culture media. The levels of endostatin in the hypoxic groups were significiently higher than the controls. The collagen ⅩⅧ mRNA expression was also different between hypoxic and normoxic groups. The expression of collagen ⅩⅧ mRNA was increased by hypoxia. The expressions of endostatin and collagen ⅩⅧ mRNA in HUVECs were significiently decreased after reoxygenation. Conclusions Hypoxia can increase the expression of collagen ⅩⅧ and endostatin in HUVECs. Hypoxia may be one of causes that influence the expression of collagen ⅩⅧ and endostatin.
