摘要
目的分析水蛭热刺激分泌物(Wp—hs)与日本血吸虫成虫可溶性物质(Sj-Ds)间的共同组分,探讨两者之间免疫交叉反应的物质基础。方法通过升高温度刺激水蛭虫体产生Wp—hs,同时收集室温放置的水蛭分泌物作为对照,用十二烷基硫酸钠一聚丙烯酰胺凝胶电泳分析Wphs与Sj—Ds之间的共同蛋白组分;将这些共同蛋白组分分别与日本血吸虫病患者混合血清、兔抗Wp—hs免疫血清、健康人混合血清进行斑点酶联免疫吸附试验和免疫印迹试验,找出具有免疫交叉反应活性的共同蛋白组分。结果Wp—hs与Sj-Ds之间存在着分子量(12.6~85.4kDa)相近的蛋白条带。Wphs抗原在21.6~95.4kDa分子量范围内与日本l皿吸虫病患者混合血清反应尤为明显,且不与正常人血清反应;Sj-Ds(40.1~89.0kDa)与兔抗Wp—hs免疫血清能较好的结合。结论Wphs与Sj—Ds之间至少存在3种具有免疫活性的共同蛋白组分。
Objective To analyze the mutual components between Whitmania pigra heat secretions(Wp-hs)and Schistosoma japonicum dissolvable substances(Sj-Ds),and explore the mechanism of the immune cross-reaction between them.Methods The heat secretions of Whitmania pigra obtained by stimulated with high temperature,and Sj-Ds were analyzed by SDS-PAGE technique,and used to react with the sera of patients with schistosomiasis,sera of healthy people,and sera of rabbits immunized with Wp-hs by Dot-ELISA and Western blot.The secretion of a Whitmania pigra at room temperature for 30 mins were collected as control.Results There were many similar molecular weight(12.6--85.4 kDa)protein bands between Wp hs and Sj-Ds,and some protein bands between 21.6-95.4 kDa from Wp-hs had intensive cross-reaction with the sera of pa tients with schistosomiasis,but no cross-reaction with the sera of healthy people.Some components of Sj-Ds whose molecular weight were from 40.1 kDa to 89.0 kDa could combine with the sera of rabbits immunized with Wp-hs.Conclusion There are at least three mutual immunocompetent protein bands with similar molecular weight between W.pigra and S.japonicum.
