摘要 【目的】研究基质金属蛋白酶-7(matrix metalloproteinase-7,MMP-7)和基质金属蛋白酶-26(ma-trix metalloproteinase-26,MMP-26)在正常子宫内膜和孕早期蜕膜组织中的表达情况。【方法】正常子宫内膜组织(n=76)按28天月经周期将其分为增殖早期(EP;n=10)、增殖中期(MP,n=11)、增殖晚期(LP,n=13)和分泌早期(ES,n=11)、分泌中期(MS,n=12)、分泌晚期(LS,n=10)以及月经期(M,n=9)。所有标本进行免疫组化和RT-PCR检测。【结果】MMP-7和MMP-26蛋白主要定位于子宫内膜腺上皮细胞。RT-PCR及免疫组化半定量分析结果表明:MMP-7高表达于分泌晚期和月经期,低表达于分泌早、中期(u=2.128,P=0.033;u=4.178,P<0.01);MMP-26的表达在增殖期逐渐上升,月经中期(LP+ES)达到高峰后(u=2.240,P=0.025;u=3.686,P<0.01)开始回落,直至分泌晚期和月经期的最低水平(u=2.172,P=0.030;u=4.146,P<0.01)。孕早期蜕膜组织(n=9)中无MMP-7和MMP-26表达。由于MMP-26表达模式和ER-α相似,【结论】MMP-7和MMP-26蛋白都选择性地表达于子宫内膜上皮部分,但表达模式并不相同。MMP-7促进子宫内膜的剥脱和重建,MMP-26则参与子宫内膜种植过程。 [Objective]To examine expression of MMPL7 and MMP-26 protein and mRNA in cycling human endometrium and early pregnancy deciduas. [Methods]Normal endometrial specimens ( n = 76) were classified according to an ideal 28-day reproductive cycle as early, mid and late proliferative phase, early, mid and late secretory phase, and menstrual phase. All samples were analyzed using Immunohistochemistry and RTPCR. Rank test was used to evaluate the significance of differences between the groups. [Results]Immunostaining localized MMP-7 and MMP-26 in epithelial glandular and luminal cells. Semi-quantifications with RTPCR and immunohistochemistry revealed maximal levels of MMP-7 in late secretory and menstrual phase whereas early and mid secretory phases had no MMP-7 signal. Expression of MMP-26 increased during proliferative Phase to a maximum at mid-cycle, then decreased to non-detectable levels in the late secretory and menstrual phases. The decidual samples tested( n=9) had no signal of MMP-7 and MMP-26. The pattern of MMP-26 expression mimicked that of ER-α. [Conclusion]MMP-7 and MMP-26 proteins are both selectively localized in the epithelial compartment of normal endometrial tissue, but their expression patterns throughout the menstrual cycle are quite different. High expression of MMP-7 in menstrual and proliferative phases suggests that MMP-7 be involved in the broken down and growing endometrium, whereas MMP-26 be involved in the implantation of endometrium since its peak expression at mid-cycle.
