摘要
目的研究肾癌细胞株中纯化的HSP70-PC体外诱导DC成熟及激活特异性抗肿瘤免疫的作用。方法采用亲和层析及离子交换层析法纯化肾癌细胞HSP70-PC,体外致敏DC,流式细胞仪检测DC表型变化,用DC体外诱导CTL产生,MTT法比较其对不同肿瘤细胞的杀伤活性。结果HSP70-PC和TNF-α均能使DC高表达分化相关抗原CD1a、CD83、HLA-DR,肾癌细胞中纯化的HSP70-PC致敏的DC体外诱导的CTL对肾癌细胞有较高的杀伤活性。结论肾癌细胞中纯化的HSP70-PC能有效地诱导DC成熟,并使其具有活化针对肾癌细胞的特异性CTL的能力。
[Objective] To investigate HSP70-PC derived from renal carcinoma ceils induces dendritic cell maturation and special antitumor immunity. [Methods] Purify HSP70-PC from renal carcinoma ceils by affinity chromatography and ion-exchange chromatography. DCs pulsed with the HSP70-PC were detected the phenotypes by flow cytometry. The cytotoxity of CTLs activated by the DCs against the different cancers cells were evaluated by MTT method. [Results] Both the HSP70-PC and TNF-α could induce DC high expression of CDla, CD83, HLA-DR, but the DCs induced by the HSP70-PC derived from renal carcinoma ceils made the CTLs activated by it had the higher cytotoxity against the renal carcinoma ceils than the other cancer cells. [Conclusion] The HSP70-PC derived from renal carcinoma cells can effectively induce DC maturation and make it possess the ability of activating the special CTLs against renal carcinoma cells.
