电针对局灶性脑缺血再灌注大鼠缺血脑区细胞黏附分子表达的影响 被引量:12

摘要 目的观察大鼠局灶性脑缺血再灌注后缺血脑区细胞间黏附分子-1(ICAM-1)、P-选择素(P-selectin)的表达和针刺对两者的影响。方法采用大脑中动脉线栓法制备MCAO缺血再灌注模型,分别于再灌注24h和针刺后取脑组织,应用免疫组化SABC方法检测ICAM-1、P-selectin的表达。结果脑缺血再灌注使缺血脑区皮质、基底节区微毛细血管ICAM-1、P-selectin表达水平增高(模型组与正常组、假手术组比较有极显著差异,P<0.01);电针可降低再灌注后升高的ICAM-1、P-selectin的表达(电针组与模型组比较有显著差异,P<0.05)。结论早期针刺治疗对脑缺血损害有效防治作用是通过对脑缺血再灌注炎性反应的免疫调节机制而实现的。 Objective: To observe dynamic changes of cell adhesion molecules ICAM-1,P-selectin protein in the ischemia brain after CI and the effect of electroacupuncture on both ICAM-1 and P-selectin. Methods: Used the suture in MCA to establish MCAO ischemia-reperfusing model. Used immunohistochemistry SAtK3(Spreptavidin-biotin-peroxidase complex) method to investigate the expression of ICAM-1, P-sdectin in rats'ipsilateral cortex and ipsilateral basal ganglia after ischemia-reperfusion and electroacupuncture. Results: The expression of ICAM-1 and P-selectin in rats' ipsilateral cortex and ipsilateral basal ganglia increased after ischemia-reperfusion. Then electroacupuncture could make the expression of ICAM-1 and P-selectin in rats' ipsilateral cortex and ipsilateral basal ganglia decrease.Conclusion: The mechanism of early acupuncture treating can effectively intervene the injury of CI, is through the immunoregulation to the inflammation reaction after cerebral ischemia.
