补肾活血法对再障小鼠骨髓基质黏附分子的影响 被引量:6

摘要 目的:观察补肾活血法对再障小鼠骨髓基质黏附能力及黏附分子CD106/CD49d的影响。方法:用免疫介导法复制小鼠再障模型,并给予补肾活血方药(参芪益肾饮),体外培养再障小鼠骨髓基质细胞,碘化丙啶染色检测其细胞周期,用流式细胞仪测定骨髓基质黏附分子表达水平。结果:再障小鼠骨髓基质细胞绝大部分处于G0/G1期,处于S期及G2+M期的细胞比例明显降低,中药组小鼠骨髓基质细胞处于S期及G2+M期的比例较模型组小鼠明显增加;模型组骨髓基质细胞黏附能力下降,中药组基质细胞黏附能力回升,与正常组无区别;模型组骨髓基质细胞CD106、CD49d表达均降低,中药组CD106、CD49d水平有所升高。结论:补肾活血法提高再障小鼠骨髓基质黏附分子表达水平,可能是其治疗再障的机制之一。 Objective : To observe the effect of Chinese herb formula with function of Strengthening - kidney - and - activating - circulation on ability of bone marrow stroma and expression of cell adhesion molecule in model of immune - mediated aplastic anemia(IMAA). Methods: Iunnunologically mediated methods were used to imitate the model of aplastic anemia in Balb/c mice. Experimented mice were divided into three groups. The marrow cell was taken after 9 to 12 days and the cell suspension was cultured in the system of Dexter's culturing. At the 28th day of experiment, the culturing cells were measured by flow cytometry. The adhesion ability, expression of vascular cell adhesion molecule(VCAM- 1) and very late antigen (VLA-4) were analyzed. Results: The adhesion ability of bone marrow stroma and expression of cell adhesion molecule( VCAM - 1, VLA -4) of IMAA mice were lower than the control group( P 〈0.01 ), Chinese herb formula could improve these indices of bone marrow stroma damage( P 〈 0.01 ). Conclusion : Strengthening - kidney - and - activating - circulation therapy can improve the expression of cell adhesion molecule( VCAM - 1 ,VLA -4) of IMAA mice, which may be one of the mechanism that these formula can cure aplastic anemia.
