旋毛虫对三硝基苯磺酸诱导肠炎小鼠Th1/Th2类细胞因子基因表达的影响 被引量:4

摘要 目的:研究旋毛虫对三硝基苯磺酸(TNBS)诱导的实验性小鼠肠炎模型Th1/Th2类细胞因子基因表达的影响。方法:雌性BALB/c小鼠随机分为50%乙醇对照组、旋毛虫(T.spiralis)应用组、TNBS诱导肠炎模型组、预先感染T.spiralis后诱导TNBS模型组,(每组小鼠取材时保证6只以上)。采用RT-PCR方法观察感染旋毛虫和未感染旋毛虫小鼠于TNBS诱导肠炎后3 d和7 d时不同基因的表达变化,包括小鼠脾脏中IFN-γ、IL-4、IL-10 mRNA和结肠中IL-10 mRNA的表达量。结果:预先感染T.spiralis后诱导TNBS模型组小鼠在造模后3 d脾脏中IFN-γmRNA的表达量与模型组比较差异无显著性(P>0.05),在造模后7 d脾脏中IFN-γmRNA的表达量低于模型组(P<0.05),IL-4 mRNA的表达量于3 d和7 d时均高于模型组(P<0.05)。预先感染T.spiralis后诱导TNBS模型组小鼠在造模后3 d和7 d脾脏和结肠中IL-10 mRNA的表达量均高于模型组(P<0.05)。结论:旋毛虫对TNBS诱导的实验性小鼠肠炎外周免疫作用机制可能通过下调炎症性肠病过度的Th1型免疫反应、上调Th2型免疫反应而实现;IL-10既作为Th2类细胞因子又作为Tr1型细胞因子对实验性小鼠的肠道局部和外周免疫均产生重要调节作用。 Obioetive To study the levels of Th1 and Th2 type cytokines mRNA in TNBS colitis without or with Trichinella spiralis (T. spiralis) infection in mice. Mothocls Female BALB/C mice were randomly divided into 4 groups: 50% Ethanol, T. spiralis only, TNBS, T. spiralis +TNBS (at least 6 in each group when mice were killed). The levels of IFN-γ, IL-4, IL-10 mRNA in spleen and IL-10 mRNA in colon in mice with colitis induced by TNBS without or with T. spiralis infection 3 and 7 d post-induction were assessed using RT PCR method. Flosults Compared with TNBS group, IFN-γ mRNA production in spleen were reduced in T. spiralis + TNBS group 7 d post-induction of colitis (P〈0. 05), IL-4 mRNA levels were increased in T. spiralis +TNBS group both 3 and 7 d post-induction of colitis (P〈0. 05). A statistically significant alteration was not detected in spleen IFN-γ mRNA level 3 d post induction of colitis (P〉0. 05). IL-10 mRNA production in colon and spleen were increased in T. spiralis + TNBS group 3 and 7 d post-induction of colitis (P〈 0.05). Oonclusion T. spiralis infection significantly attenuates TNBS induced colitis in mice. The peripheral immunologic mechanism is that T. spiralis can down-regulate strongly Thl type immune response of colitis and up-regulate Th2 response. At the same time, IL-10 as Th2 and Tr1 type cytokine has an important effect on colonic immunity and general immunity of TNBS induced mice.
