摘要 1,3-丙酮二羧酸二乙酯与硫和丙二腈环合后,与氯乙酸乙酯缩合得5-[N,N-二(乙氧羰甲基)氨基]-4-氰基-3-乙氧羰甲基-2-噻吩甲酸乙酯,再用氢氧化锶水解、成盐制得抗骨质疏松症药物雷尼酸锶,总收率34%。 Strontium ranelate, a bone formulation stimulant, was synthesized from diethyl 1,3-acetonedicarboxylate by cyclization with sulfur and malononitrile, condensation with ethyl chloroacetate to give 5-[N,N- bis (ethoxycarbonylmethyl) amino] -4-cyano-3-ethoxycarbonylmethyl thiophene-2-carboxylic acid ethyl ester, which was subjected to alkaline hydrolysis with strontium hydroxide in 34% overall yield.
