摘要 用均匀设计法优化了多杀菌素发酵培养基的配方,同时探讨了前体物质对发酵效价的影响。结果得到优化培养基组成(g/L):葡萄糖37.8,糊精42.0,棉籽粉22.3,脱脂奶粉9.0,CaCO33.0。发酵30h后添加10mmol/L的正丁醇效果最佳,优化后多杀菌素的效价为394.1μg/ml,比优化前提高了56%。 Uniform design method was employed to optimize the fermentation medium. Meanwhile, the effects of precursors on the biosynthesis of spinosad were also studied. Components of the optimized medium (g/L)were 37.8 glucose, 42.0 dextrin, 22.3 cottonseed flour, 9.0 skimmed milk powder and 3.0 calcium carbonate. The fermentation potency of spinosad could reach to 394.1μg/ml when flask fermentation by Saccharopolyspora spinosa sp-29 was carried out with shaking in the optimized medium feeding 10mmol/L n-butanol at 30h, which had increased by 56%.
