摘要
将(亚洲棉×草棉)F1进行染色体加倍,再与(陆地棉×海岛棉)F1进行杂交,产生亚洲棉、草棉、陆地棉和海岛棉的四元杂种.四元杂种形态特征综合有亚洲棉、草棉、陆地棉和海岛棉的一些性状,总体植株形态似陆地棉.SSR分子标记分析结果表明,45对SSR引物中,具多态性的引物共30对,共检测到99条清晰的多态性带,平均每个SSR座位可检测到3.3个.四元杂种具有4个亲本的特征谱带,从分子水平上证明四元杂种具有4个亲本的遗传物质,其中亚洲棉特征带占20.8%,草棉特征带占22.2%,陆地棉特征带占15.3%,海岛棉特征带占13.9%,而四元杂种特有带占27.8%.四元杂种与4个亲本(亚洲棉、草棉、海岛棉和陆地棉)之间的遗传相似系数分别为0.500、0.444、0.389和0.444.研究结果表明SSR分子标记适用于棉花遗传亲缘关系和远缘杂种的鉴定,结果准确可靠;四种栽培棉种之间的亲缘关系相对较近,可以通过遗传重组产生综合有4个栽培棉种性状的棉花新种质.
The quadrispecific hybrid F1 ( G. a rboreum , G. herbaceum , G. h irsutum and G. ba rbadense ), was obtained by crossing the doubled (G. a rboreum × G. herbaceum ) F1 with ( G. h irsutum × G. barbadense ) F1 , It had some morphological characters from its four parents, but the whole plant looked like that of upland parent. The results of SSR analysis showed that there were 30 pairs of poly-morphological primers for total 45 pairs of SSR ones, and 99 polymorphism bands were obtained, with an average number of 3.3 for each SSR locus. The quadrispecific hybrid F1 had the characteristic bands of four parents and accordingly had its parent genetic materials from SSR molecule marker analysis. Among the SSR markers in the quadrispecific hybrid F1,20.8% of them come from G. arboreum, 22.2% from G. herbaceum, 15.3% from G. hirsutum, and 13.9% from G, barbadense. Other 27.8% markers were the new and peculiar ones for the hybrid. Jaccard' s genetic similarity coefficients among quadrispecific hybrid F1 and its four parents G. arboreurn, G. herbaceum, G. hirsutum and G. barbadense were 0. 500,0. 444,0. 389 and 0. 444 respectively. According to this result, it shows that SSR molecular marker is a feasible method in studies of genetic relationship and identification in interspecific cotton hybrids. The genetic relationship among the four cultivated cotton species is near relative, and it is possible to produce a new germplasm recombined outstanding characters from four cultivated cotton species via the interspecific hybridization.
