摘要
利用随机扩增多态DNA(RAPD)标记,对国家二级重点保护植物香果树(Emmenopterys henryi)的遗传多样性和遗传分化进行了分析.12个随机引物对9个香果树居群173个个体的DNA样品进行扩增,共检测到182个扩增条带,其中100条为多态条带,多态位点百分率(PPB)为54.95%;香果树总的Shannon多样性指数(I)为0.2838,Nei指数(h)为0.1900,表明种水平的遗传多样性较高.而居群水平的遗传多样性较低,PPB平均为16.48%,I平均为0.0914,h平均为0.0622.AMOVA分子差异分析表明香果树居群间遗传分化程度高,74.76%的变异存在于居群间,25.24%的变异存在于居群内.香果树居群间的基因流很低,为0.2434.9个居群间的平均遗传距离为0.1653,遗传距离与居群间的地理距离存在显著的相关性.利用算术加权平均数法(UPGMA)对香果树居群进行聚类,结果9个居群可分成浙江省内和省外两大类群.
Genetic diversity and genetic differentiation of Emmenopterys henryi, a national Class Ⅱ protection plant, were analyzed by using random amplified polymorphie DNA (RAPD) marker, 9 populations of Emmenopterys henryi with 173 individuals were amplified with 12 random primers and total 182 amplified bands were produced, Among which, 100 bands were polymorphie and the percentage of polymorphie bands (PPB) was 54.95%. The genetic diversity at species level was relatively high, Shannon information index (Ⅰ) was 0. 2838 and Nei's gene diversity (h) was 0. 1900, The genetic diversity at population level was relatively low. The mean of PPB was 16.48%, the mean of Ⅰ was 0. 0914 and the mean of h was 0. 0622, Analysis of molecular variance (AMOVA) demonstrated that the genetic differentiation among population was high, Variation among population occupied for 74.76%, while that within population occupied only for 25.34%. The gene flow among population was very low down to 0. 2434, The mean genetic distance of 9 populations of Emmenopterys henryi was 0. 1653, There was significant correlation between genetic distance and geographical distance, Using unweighted pair group method with arithmetic average (UPGMA), 9 populations of Emmenopterys henryi were clustered into two groups, inner-province group and outer-province group.
