摘要
目的:研究体外培养的皮质神经元在谷氨酸作用下,其凋亡诱导因子(AIF)向细胞核转位的情况。方法:用谷氨酸诱导体外培养的皮质神经元凋亡,用免疫荧光指示热休克蛋白-60(HSP-60)和AIF的亚细胞分布,4,6-二脒-2苯-基吲哚(DAPI)显示细胞核,并根据DAPI显示的细胞核形态及其位置判断细胞凋亡。结果:所有神经元都表达AIF。正常情况下AIF分布在线粒体;经谷氨酸处理后AIF从线粒体释放,并可转位至细胞核,致部分细胞可能发生凋亡。结论:谷氨酸可引起AIF从线粒体释放并向细胞核转位,同时可诱导体外培养的原代皮质神经元凋亡。
Objective:To investigate the apoptosis-inducing effects of glutamic acid on primary cultures of rat cortical neurons, and on the translocation of apoptosis-inducing factor (AIF) from mitochondriato nuclei. Methods:The apoptosis of cortical neurons was induced by glutamic acid. The subcellular distribution of AIF and heat shock protein 60 (HSP-60) indicating the location of cytosol was revealed by immunofluorescence, with 4,6-diamidino-2-phenylindole (DAPI) indicating nuclei. Cellular apoptosis was judged by the nuclear modality and the position relative to cytoplasm showed by DAPI. Results:Almost all neurons expressed AIF. AIF located in mitochondria for the normal neurons and in some extent translocated from mitochondria to nuclei in part of the neurons, after treated with glutamic acid. Conclusion:Glutamic acid could bring about the translocation of AIF from mitochondria to nuclei and induce cortical neurons to undergo apoptosis.
