摘要 目的:探讨雌激素受体在大鼠全层皮肤缺损创面愈合中的表达情况及相关的分子机制。方法:Wistar雄性大鼠30只,在背部制成全层全损皮肤创伤愈合模型,分别于伤后1、3、5、7、15和21 d取材。常规组织学观察和免疫荧光染色,观察皮肤愈合过程与雌激素受体和原癌基因c-fos的表达变化规律。结果:损伤早期(1-3 d)ER-β和c-fos的表达逐渐增强,5 d时,ER-β和c-fos阳性表达进一步增加,但主要集中在真皮层,表皮层的表达较少。伤后7 d,在创缘部分角质形成细胞开始表达ER-β和c-fos。15 d后,ER-β和c-fos的表达仍维持在较高水平,创缘区域的表达尤其明显。伤后21 d,两者的表达有所减弱,但在愈合部位的表皮层角质形成细胞和真皮层血管内皮细胞仍有较强的表达。结论:雌激素受体系统参与全层皮肤缺损的愈合过程,影响组织的肉芽组织形成,再上皮化以及愈合后的塑性,可能与原癌基因c-fos蛋白的表达有关。 To investigate the expression of estrogen receptor beta ( ER - β) in human fetal skin, and elucidate its role during skin wound healing. Methods: Thirty Wistar diabetic rats were used in this study. Four full - thickness skin wounds were incised in the back of each rat. The wound healing was observed by measuring the wound area from 1 to 21 days. The expressions of ER - β and c - fos were detected with immunofluorescence staining technique. Results: From 1 to 3 days after injury, the expressions of ER - β and c - fos were mainly restricted to the dermal fibroblasts, inflammatory cells and vascular endothelial cells. Five days after injury, the expressions were increased, and a few of immuno - reactive positive cells were detected in keratinocytes of wound edges. Seven days after injury, both ER-β and c -fos were strongly expressed not only in the dermis but also in epidermis. Fifteen days after injury, the expressions maintained a highly level, especially in wound sites. After 21 days, the ER-β and c -fos were weakly expressed, but were still expressed in keratinocytes of epidermis and vascular endothelial cells of dermis. Conclusion: These results provides further evidence that estrogen receptors involve in the procession of skin injury and repair, suggesting that estrogen system may play an important role during wound healing via oncogene c -fos proteins.
