摘要
本文研究了单键DNA分子(ssDNA)在石墨电极上的固定方法,采用核酸分子杂交技术,使具有电化学活性的米托蒽醌(MX)嵌入DNA分子双螺旋结构的碱基对中,在电极上形成dsDNA-MX层,通过伏安法研究DNA分子和MX相互作用的电化学行为,并且根据嵌入剂MX的氧化峰电流计算出DNA分子与MX的表观缔合常数K=8.4×104(mol/L)-1,结合位点的大小n=5。
A synthesized 75-mer single -stranded DNA- 1 (ssDNA-1 ) was covalently immobi lizccl onto an aminated graphite electrode surtace . At 42℃' in 2 × SSC hybridization buffer solution containing 1. 0mg/L complementary ssDNA-2 and 1. 0 ×10- 3 mol /L mitoxanthrone (MX ). the ssDNA-1 immobilized onto the electrode hybriclizes with ssDNA-2 forming dou ble stranded DNA (dsDNA ). MX was capable of intercalating between base pairs of the dsl)NA layer on the electrode . The electrochemical behaviour of the dsDNA and MX intercala tion was studied by electrochemical signals of MX. The apparent association constant of MX-DNA is 8. 4 × 104 (mol/L ) - 1, and the binding size is 5.
