摘要
目的:观察丹参注射液对急性肺损伤(ALI)大鼠的治疗作用及其机制。方法:Wistar大鼠30只,被随机分为正常组、模型组及丹参干预组,每组10只。采用“二次打击”的方法复制AL I大鼠模型,即从尾静脉注入油酸(OA)0.2ml/kg,4h后再从尾静脉注入脂多糖(LPS)2mg/kg。注射OA前1h经腹腔注射丹参注射液12ml/kg作为干预措施。观察支气管肺泡灌洗液(BALF)中蛋白含量、肺脏的大体及光镜下病理改变,以及肺组织湿/干重(W/D)比值,并测定肺组织中一氧化氮(NO)、丙二醛(MDA)、超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)的变化。结果:“二次打击”法可成功复制出ALI大鼠模型。与模型组相比,丹参干预组BALF中蛋白含量、肺组织W/D比值和MDA、NO水平均明显降低(P均<0.01),而肺组织SOD、GSH-Px活性则明显升高(P均<0.01),并且丹参干预组可明显减轻“二次打击”所致的肺组织病理改变。结论:丹参注射液通过抗氧化作用、调整氧化/抗氧化平衡,对ALI大鼠具有较好的保护作用。
Objective: To observe the effect and mechanism of radix salviae miltiorrhizae (RSM) injection (丹参注射液) on rats with acute lung injury(ALI) induced by "two hits". Methods: Thirty Wistar rats were randomly divided into three groups : normal control group, model group and RSM treatment group (each n= 10). The ALI model was induced by "two hits" : 0.2 ml/kg oleie field (OA) was injected into one tail vein, and 2 mg/kg lipopolysaeeharide (LPS) was injected into the vein 4 hours later. One hour before the injection of OA, 12 ml/kg RSM injection was injected into the peritoneal cavity as the pretreatment measure. Then, the content of protein in bronehoalveolar lavage fluid (BALF) and the general and pathologleal changes of rat lungs were observed, and the wet and dry weight ratio (W/D) of lung was calculated. In addition, the concentrations of nitric oxide (NO) and malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH- Px)in lung tissues were measured by the method of eolorimetry. Results: ALI was obviously observed in the model group. In model group, the activities of SOD and GSH -Px were decreased significantly (both P〈 0.01), however, the content of protein in BALF, the MDA, NO contents in lung tissues, and W/D of lung were increased markedly compared with those of RSM group (all P〈0.01), and the pathological changes of the lungs induced by "two hits" were also ameliorated in RSM treatment group. Conclusion: RSM injection has protective effect on ALI in rats by anti-oxidation and adjusting oxidation/anti-oxidation balance.
