Preparation of sparfloxacin liposome by ethanol injection

摘要 制备司帕沙星脂质体,单因素考察优化了制备工艺和处方,以提高脂质体的包封率.采用乙醇注入-高压均质法制备了司帕沙星脂质体,以透析-高效液相色谱法测定了含量和包封率.实验结果表明,司帕沙星脂质体的平均粒径为22.1 nm,包封率为47%左右.乙醇注入-高压均质法适用于制备司帕沙星脂质体,透析-高效液相色谱法操作简单、准确、重复性好,可用于测定司帕沙星脂质体的含量和包封率. With the aim to prepare sparfloxacin liposome and to improve entrapment efficiency, the main factor affecting the quality of sparfloxacin liposome in the preparation was studied by single factor exploration. Sparfloxacin liposome was prepared by ethanol injection and high homogenizer methods. Sparfloxacin liposome and its entrapment efficiency were determined by dialysis-HPLC. The results revealed that the mean size of liposome was 22. 1 nm and entrapment efficiency was about 47%. It was viable to prepare liposome such as Sparfloxacin by ethanol injection and high homogenizer methods. Dialysis-HPLC method was convenient, accurate and suitable for the determination of Sparfloxacin and the entrapment efficiency in Sparfloxacin liposome.
