摘要
目的探讨过敏性疾病主要变应原之一德国小蠊Bla g 2的定位。方法制作德国小蠊石蜡切片,在本室克隆、表达和纯化出Bla g 2蛋白的基础上,按常规方法免疫小鼠,分离血清用作一抗,荧光染色后在光镜下观察Bla g 2在虫体内的定位。结果用rBla g 2免疫小鼠血清孵育的切片中,中肠肠腔上皮细胞和肠内容物显示强阳性,免疫小鼠血清孵育的粪便颗粒也显示阳性结果。而其生殖、排泄系统等脏器均呈阴性反应。结论本研究发现德国小蠊II类变应原Bla g 2存在于中肠肠腔上皮组织、肠内容物和粪便颗粒中,对进一步进行德国小蠊特异性抗原的分离、纯化及疫苗的研究提供了理论参考。
The present study was attempted to study the localization of the German cockroach major allergen Bla g 2 in the paraffin section with Blattella germanica by using the monospeeifie antisera against Bla g 2 allergen. Cockroaches were embedded on paraffin section and serial sections were prepared for light microscopic examinations. These sections were examined by immunohistoehemieal staining with the monospeeifie antisera against Bla g2 allergen previously prepared in our laboratory. For immunostaining, the cockroach sections were firstly incubated with mouse anti-rBlag 2 antiserum and then the biotin-labeled anti-mouse IgG antiseum was used as the secondary antibody. It was showed that positive reaction of staining with caballing of the epidermal cells in the midgut and intestinal contents as well as in the fecal pellets could be demonstrated, but no other internal organ was positively labeled. These results indicate that the German cockroach major allergen Bla g 2 is most likely derived from the digestive tract of cockroaches.
