用PCR-限制性片段长度多态性分析法检测游泳池肉芽肿中海分枝杆菌 被引量:7

摘要 目的:探讨用PCR-限制性片段长度多态性(RFLP)分析法快速检测游泳池肉芽肿组织中海分枝杆菌。方法:对疑诊为游泳池肉芽肿患者5份皮损组织DNA进行PCR扩增,扩增产物分别用BstEⅡ和HaeⅢ两种限制性内切酶进行酶切,再用琼脂糖凝胶电泳进行RFLP分析,并与培养结果进行比较。结果:5份标本中均检测出海分枝杆菌,与培养结果一致。经过3~7个月治疗,4例患者治愈,1例患者好转。结论:用PCR-RFLP可以快速鉴定海分枝杆菌,有利于该类疾病的早期诊断和及时治疗。 Objective: To establish a rapid approach for detection of M. marnium from lesions of patients with suspected M. marinum infection. Methods: Specimens were obtained from 5 patients with swimming pool granuloma (SPG). DNA was extracted and PCR was performed, and PCR products were identified by restriction fragment length polymorphism (RFLP) analysis. The candidate gene was heat shock protein 65 kD. Results: M. mozinum was detected in all specimens from patients with SPG, and was consistent with bacteriological identification. The lesions disappeared within 3 to 7 months after treatment. Conclusions: PCR-RFLP is a sensitive, specific and rapid method in identification of mycobacterium. Application of this method will be helpful for early diagnosis of mycobacterial skin infections.
