摘要
以邻氨基苯酚、邻硝基苯酚和丙烯醛为原料,在乙酸-盐酸水溶液中,用Skraup法合成了8-羟基喹啉。优化的反应条件为:n(邻氨基苯酚):n(邻硝基苯酚):n(丙烯醛):n(乙酸)=1:0.5:1.8:2.0,90~100℃反应5h,经中和、蒸馏得到8-羟基喹啉,收率136%(以邻氨基苯酚计)。通过元素分析、红外光谱和核磁共振对8-羟基喹啉的结构进行了表征。
8-Hydroxyquinoline, useful as an important medicine intermediate, was synthesized by Skraup method from o-aminophenol, o-nitrophenol and acrolein in the acetic acid and HC1 aqueous so- lution under the optimal conditions: molar ratio of o-aminophenol : o-nitrophenol " acrolein " acetic acid= 1.0 : 0.5 : 1.8 : 2.0, and reaction at 90 - 100 ℃ for 5 h to give 8-hydroxyquinoline with yield of 136% (calculated based on o-aminophenol added)after workup by neutralization and distillation. 8- Hydroxyquinoline was confirmed by elemental analysis, IR, and ^1H NMR.
