摘要 目的建立粒细胞减少小鼠大腿动物感染模型。方法ICR小鼠腹腔注射不同剂量环磷酰胺,取外周血行白细胞分类计数;予小鼠大腿接种金黄色葡萄球菌标准菌ATCC29213,观察24h存活率、外周血粒细胞及肌肉菌量变化。结果环磷酰胺(150+100)mg·kg-1剂量组粒细胞数达到预期水平,连续4天低于100个.μL-1;对数生长期菌液百倍稀释后浓度约1×107CFU·mL-1;接种24h后,小鼠存活率75%,中性粒细胞数均显著升高;26h后,大腿组织菌量为3.55×108CFU。结论本文建立的粒细胞减少小鼠大腿感染动物模型特征稳定。 Objective To establish a neutropenic -mouse thigh with S. aureus infection model. Methods ICR mice were given different doses of cyclophosphamide (CPA) intraperitoneally. The change of leukocytes and neutrophils in peripheral blood were observed. Inoculum of S. aureus ATCC 29213 were injected into the thighs. The distribution of death of mice were recorded as well as the change of neutrophils and bacterial counts. Results The neutrophils counts in ( 150 + 100) mg · kg^-1 CPA doses group were less than 100 count ·μL^-1 for4 days. The 1:100 dilution of bacterial suspension in logarithmic phase was about 1 × 10^7CFU · mL^-1. The survival rate of mice was 75%. The neutrophils counts increased after inoculation. The number of CFU in thighs after 26 h of inoculation is 3.55 ×10^8 CFU. Conclusion The neutropenic - mouse thigh infection model in this study has steady characters of animal model.
