胃溃疡复发模型大鼠胃组织蛋白质组的动态变化及健胃愈疡颗粒干预作用 被引量:4

Dynamic changes of proteome in the process of gastric ulcer recurrence and intervention of Jianweiyuyang granule in rats

摘要 [目的]通过胃溃疡(GU)复发大鼠胃组织蛋白质差异显示及健胃愈疡颗粒(JWYY)干预,寻找JWYY抗GU复发的相关蛋白。[方法]采用乙酸及白细胞介素1β(IL-1β)复制GU复发动物模型,分别于造模后第8、16、92天,对大鼠GU复发过程进行动态观察,并同时给予JWYY干预,收集各时间点大鼠胃组织,提取胃组织总蛋白质,分别进行双向凝胶电泳(2-DE)分离I、mage Scanner扫描、PDQuest行差异蛋白质分析、凝胶考染,显示差异蛋白质。[结果]模型复发组大鼠出现生命活动整体反应(如体重、活动度等)下降;在各组考染凝胶图谱上,可辨识的蛋白质斑点达900-1 300个;与模型复发组相比,JWYY干预后,其蛋白质表达在不同时间点均有差异。[结论]JWYY抗GU复发的作用机制是多方面的,其干预作用与多种蛋白质相关。 [Objective]To demonstrate the differences of gastric tissue proteome in gastric ulcer recurrence rats and the intervention effect of JWYY Granule. [Methods] The animal model of gastric ulcer recurrence was duplicated by acetic acid and IL-1β,on the 8^th, 16^th, and 99^nd day respectively to observe the process of the gastric ulcer recurrence dynamically in rats and therapeutic intervention of JWYY. The gastric tissue was collected at each time point, total protein of gastric tissue was extracted and separated by 2-DE (2 dimensional electrophoresis) and scanned by Image Scanner and the different proteins were analysesed by PDQuest and the differential protein was detected among varied groups by Coomassie brilliant blue stain. [Results]The integral response of vital movement such as body weight and activity declined in the gastric ulcer recurrence rats;According to gel maps among varied groups, approximately 900 to 1300 protein spots were visible. And special proteins varied at different time point with intervention of JWYY compared with the group of recurrence model. [Conclusion]There are multi-aspects of the mechanism to prevent gastric ulcer recurrence with JWYY Granule and its intervention effects could be related to varied proteins.
