摘要
应用Hungate厌氧技术,从大庆油田常规水驱采出液中分离到一株硫酸盐还原功能菌株A9,进行形态、生理生化、16S rDNA以及16S-23S rDNA间隔区序列鉴定以及生长因子研究.该菌株为G+,短杆状,能运动,具有硫酸盐还原功能,产H2S,兼性厌氧;能以水溶性聚丙烯酰胺为唯一碳源;A9菌株与Anaerofilum pentosovorans(AP716816S)的相似性水平达98%,初步鉴定可能为Anaerofilum属的新种,暂时命名为Anaerofilum pentosovoransA9.生长因子及其正交试验结果表明,pH、温度都达到极显著的水平,聚合物影响达到显著水平;菌株最适生长pH为8.0,喜中性偏碱环境;最适温度为40℃,属于嗜中温菌;聚合物质量浓度为600 mg/L生长较好,菌株对降低聚合物黏度效果明显;总矿化度最适生长范围为1.5×103-2.5×104mg/L.
Using the Hungate anaerobic technology, a function bacteria of sulfate reducing bacteria named A9 was separated from the out - liquid of the forth general water - drive factery of Daqing Oil Field. The identif- ying of morphological, physiological, biochemical, 16S rDNA and 16S - 23 S rDNA Intergenic Spacer sequence as well as the research of its Growth Factor were carried out. This bacteria is G ^+ , short bacillus and athletic. It can reduce the sulfate and bring out HES. It is intersexual anaerobic. It can use the hydrosoluble polyacrylamide (HPAM) as the only carbon source; the comparability between the bacteria A9 and Anaerofilum pentosovorans (AP716816S) reaches to 98% ; the primary identification judges that it may be a new species of the Anaerofilum category. It is named as Anaerofilum pentosovorans A9 provisionally. The experiments of growth factor and orthogonality show that the pH value and tempreture reach the prominent level, and the effect of polymer also reaches the prominent level. The best pH value for it is 8.0, and it likes the litmusless or alkalescent circumstance. Best tempreture for it is40℃. It is the middle - tempreture bacteria. When the concentration of polymer is 600 mg/L, the A9 bacteria grows well and its effect on reducing the polymer viscidity is prominent. Best total mineralization range for growing is 1.5 × 10^3 -2. 5 ×10^4 mg/L.
