细胞壁中过氧化物酶的分布对杨树木质化过程的影响 被引量:7

摘要 本文通过二氨基联苯胺(DAB)细胞化学染色方法,利用透射电镜(TEM),研究毛白杨(Populus tomentosaCarr.)木质部细胞分化过程中过氧化物酶(POD)在细胞壁上的分布,并讨论了POD的分布位置与木质素沉积之间的关系。形成层细胞阶段和伸展生长阶段,POD主要分布在径壁上,尤其是细胞壁角隅处和初生纹孔场附近的细胞壁。在次生壁S1层的沉积开始后,胞间层的POD反应强烈。随后,POD的分布由胞间层的角隅处向S1层快速推进。但整个过程中,POD活性的增加并不是由胞间层依次向次生壁各层推进。与木质化过程的比较结果表明,POD参与了细胞壁的木质化过程,但次生壁中木质素的沉积位置并不与POD活性位置一一对应。这说明不同时期和不同位置分布的POD是具有不同功能的同工酶组份,不能将细胞壁上木质素的沉积位置与POD的分布简单地对应。 In this paper, the relationship between lignification progress and peroxidase (POD) locations in secondary xylem cell of Populus tomentosa Cart. was analyzed on the basis of cytochemistry localization and TEM observation of POD. POD was mainly distributed in the radial walls of the cambial cells and expanding xylem cell, especially at the comer of cell wall and cell wall close to the primary pit field. POD was occurred apparently at the middle lamella, and then their signals extended rapidly from the middle lamella at the coll wall comer to S1 layer with S1 layer deposition. But the increase of POD activity did not occur from S1 to S2 in turn. The present results indicated that POD had participated in the lignification. However, the distribution of POD was not same as the regions of lignin deposition completely. This therefore suggested that it was probably different components of POD isozyme with specific functions that distributed within the different cell regions at different stages in differentiation process.
