摘要
目的建立增加骨密度动物模型,并考核该模型应用于保健食品的筛选与评价的效果。方法实验设低、中、高三个剂量组,另设一个低钙对照组和一个碳酸钙对照组。观察包括生长发育、骨钙含量、骨密度及钙表观吸收率等相关指标。结果高剂量组钙的表观吸收率与相同水平的碳酸钙对照组比较皆明显增加,差异有非常显著性意义(P<0.01),表明受试样品与碳酸钙标准一样皆可作为补钙剂;各剂量组动物右侧股骨重量与低钙对照组比较增加,差异均有非常显著性意义(P<0.01),而高剂量组动物股骨重量与相同水平碳酸钙对照组比较未见显著降低;各剂量组动物左侧股骨骨密度与低钙对照组比较增加,差异均有非常显著性意义(P<0.01),但高剂量组与相应碳酸钙对照组比较降低,且差异有显著性意义(P<0.05),故不能判定骨密度结果阳性;各剂量组动物右侧股骨钙含量与低钙对照组比较增加,差异有非常显著性意义(P<0.01);高剂量组动物右侧股骨钙含量与相同水平碳酸钙对照组比较未见显著降低,表明该受试样品骨钙含量结果阳性。根据《保健食品检验与评价技术规范》(2003年版)中增加骨密度功能检验方法判定标准,试验时间共30d,其动物模型评价受试样增加骨密度功能阴性,但可作为补钙剂使用。结论增加骨密度动物模型建立成功,可应用于保健食品增加骨密度功能的评价。
Objective To establish increasing bone density animal model and use in screening and assessment of healthy foods. Methods There were five experiment groups including low, middle, high dosage groups, low calcium control and calcium carbonate control groups. The observational indexes included growth and development, bone calcium, bone mineral density and apparent calcium absorption rate. Results 1 .Apparent calcium absorption rate in the high calcium dose group was significantly higher than that of carbonate control ( P 〈 0.01 ). 2. The weight of right thigh bone in all dosage groups increased significantly as compared with that of low calcium control group ( P 〈 0.01), but there no significant difference was observed between high dosage group and carbonate control group. 3. The bone mineral density of the left thigh bone increased significantly as compared with that of low calcium control group (P 〈 0.01 ), but significantly reduced as compared with that of carbonate calcium group (P 〈 0.05 ). Thus the result of the bone mineral density was negative. 4. Bone calcium in all dosage groups were significantly lower than that of low calcium group (P 〈 0.01), but there was no significant difference between high dosage group and calcium control group, the result was positive. Above all, based on the criterion of testing and evaluating health foods(2003), the sample can be used as calcium supplement, but the function of increasing bone mineral density was negative. Conclusion This animal model can be used for evaluating the function of healthy foods capable of increasing bone density.
