摘要
目的研究组织工程皮肤中胶原合成及分泌情况,并与正常皮肤对照。方法采用苦味酸-天狼星红染色,偏振光显微镜观察,Photoshop半定量分析,观察Ⅰ、Ⅲ型胶原的含量。结果正常皮肤和组织工程皮肤的真皮中都有Ⅰ、Ⅲ型胶原,在正常皮肤的真皮中Ⅰ、Ⅲ型胶原含量分别是98.51±11.28、33.93±3.90;培养成熟的组织工程皮肤中Ⅰ、Ⅲ型胶原含量分别是91.42±3.82、30.75±2.29,二者相比差异无统计学意义(P>0.05)。组织工程皮肤培养8、15、30d,组织内Ⅰ型胶原分别为50.75±4.22、63.57±6.87、91.42±3.82,Ⅲ型胶原分别为16.90±1.64、23.65±4.08、30.75±2.29,组间比较差异有统计学意义(P<0.01)。结论(1)培养成熟的组织工程皮肤与正常人皮肤中胶原含量及分布无明显差异;(2)组织工程皮肤不同的培养时期胶原的含量及种类有明显差异。
Objective To study the synthesis and excretion of the tissue engineered skin, also to compare the variability of the tissue engineered skin with the normal skin. Methods Picric-sirius red staining was used. The percentage of the stained area in the tissue was measured with a computerized image analysis system (Photoshop). Results There were typeⅠ、Ⅲ collagen in tissue- engineered skin and the normal skin. The percentages of type Ⅰ、Ⅲ collagen in the normal skin were 98.51± 11.28 and 33.93±3. 90, the percentages of type Ⅰ、Ⅲ collagen in the tissue engineered skin were 91.42±3.82 and 30.75±2.29 . There was no signifi- cant difference in the tissue engineered skin and the normal skin(P〉0.05). The percentage of type Ⅰ、Ⅲ collagen in the tissue engineered skin was detected at 8, 15, 30d. The percentages of tyep Ⅰ collagen were 50.75±4.22 ,63.57±6.87,91.42±3.82 respectively. The percentages of type Ⅲ collagen were 16.90± 1.64,23.65 ± 4.08,30.75 ± 2.29 respectively. There was significant difference among three tissues engineered skin(P〈0.01). Conclusion (1)There is no significant difference in the percentage and distribution between type Ⅰ、Ⅲ collagen in the tissue engineered skin and normal skin. (2) There is significant difference among three tissues engineered skins.
