子宫颈部肿瘤中DNA甲基化转移酶-1表达和P16/MGMT基因启动子甲基化的临床意义 被引量:8

摘要 目的探讨子宫颈部肿瘤中Dnmt-(1DNA甲基化转移酶-1)表达和P16/MGMT基因启动子甲基化的临床意义。方法用MS-PCR(Methylation Specific-PCR)和免疫组织化学染色方法研究Dnmt-1蛋白和P16/MGMT基因启动子甲基化在117例子宫颈部良、恶性病变组织中的表达意义。结果Dnmt-1在浸润性鳞状上皮癌和腺癌中的表达明显高于慢性宫颈炎和宫颈上皮内瘤变。P16/MGMT基因启动区在慢性宫颈炎中全部表现为非甲基化,而在宫颈上皮内瘤变、浸润性鳞状上皮癌和腺癌中则表现为很高的异常甲基化水平。在子宫颈腺癌中,P16/MGMT基因启动子甲基化率在Dnmt-1表达阳性组明显高于Dnmt-1表达阴性组。结论Dnmt-1蛋白表达和P16/MGMT基因启动子甲基化在宫颈癌进展过程中的早期就起着非常重要的作用,而且Dnmt-1蛋白表达和P16/MGMT甲基化检测对于恶性肿瘤的诊断有着重要的辅助意义。 [ Objective] To investigate the significance of Dnmt-1 expression and P16/MGMT aberrant methylation in uterine cervical cancer. [Methods] Dnmt-1 expression and P16/MGMT aberrant methylation were detected using immunohistochemistry and MS-PCR (Methylation Specific-Polymerase Chain Reaction) in 117 of uterine cervical lesions. [Results] Dnmt-1 expression rate was significantly higher in invasive SCC (Squamous Cell Carcinoma) and than in cervicitis and CIN (Cervical Intraepithelial Neoplasm). The rate of P16/MGMT aberrant methylation was higher along with the increased Dnmt-1 expression in uterine cervical ad [ Conclusions] Dnmt-1 protein and P16/MGMT aberrant methylation has important roles in the early progress of uterine cervical cancer. And the detection of Dnmt-1 protein expression and P16/MGMT aberrant methylation is helpful for the diagnosis of uterine cervical cancers.
