摘要 目的观察睾丸支持细胞(sertoli cells,SCs)与骨髓间充质干细胞(mesenchymal stemcells,MSCs)共培养时SCs对MSCs扩增的影响。方法取雄性SD大鼠睾丸,分离出SCs,取SD大鼠,分离出骨髓MSCs,将两种细胞用"三明治"式的方法共培养7d。结果共培养组的MSCs前四天生长较对照组快,第5天无明显差异,第6和第7天细胞数量迅速减少。结论在体外共培养的早期,SCs可以显著促进MSCs的生长,但在后期则反而抑制MSCs的生长,具体原因有待进一步研究。 Objective To investigate the amplification of MSCs co-cultured with the SCs in vitro. Methods SCs were isolated from 12-16 day-old SD rat testes and MSCs were separated from adult SD rat bone marrow. Two kinds of cells were co-cultured for 7 days by a method of just like "sandwich". Results The MSCs in co-culture group proliferated faster than that in the control group from the 1st day to the 4th day. On the 5th day, there was no difference between coculture group and control group. But on 6th and 7th days, the MSCs amount of the co-culture group was decreased sharply. Conclusion In the early period of co-culturing in vitro, SCs may remarkably promote the amplification of MSCs, but in later period SCs suppress the proliferation of MscS.
