摘要 目的 探讨HLA-B27基因检测的临床应用效果。方法 对100例临床送检疑似强直性脊柱炎(ankylosing spondylitis,AS)病例(观察组)及30例经体检合格的献血员(对照组),采用聚合酶链反应-序列特异引物法(PCR-SSP)进行HLA-B27基因检测,并与流式细胞术检测结果比较。结果 在100例疑似AS病例中,应用PCR—SSP方法检出HLA-B27基因69例,未检出HLA-B27基因31例,30例献血员均未检出HLA-B27基因;用流式细胞术检测,得出与PCR-SSP法完全相同的结果。结论 HLA-B27检测PCR-SSP法和流式细胞术各有特点:PCR-SSP法具有结果准确、成本低、标本可长期保存,便于进行实验室间质量评价等特点;流式细胞术具有简便快捷、自动化程度高、结果判断客观性强等特点。 Objective To investigate the effect of the clinical application gene of detection on HLA-B27 gene. Hethod The human leukoeyle antigen B27 gene with PCR-SSP technique were detected for one hundred cases which were doubtful as the illness of AS (The Observed Team), as well as 30 blood samples from healthy persons (The Compared Team). And the result of this method were compared with the testing outcome by the flow cytometer. Result There were 69 out of 100 cases of doubtful AS patients, in which HLAB27 was detected by using the method of polymerase chain reaction (PCR-SSP). On the other hand, HLA-B27 gene was not found in the other 31 cases. HLA-B27 in the 30 blood samples from the healthy persons was total negative. This result was completely matched with the outcome by the flow cytometer. Conclusion Applying PCR-SSP technique to detect the HLA-B27 gene has its own feature, and also the method of flow cytometer. The advantage of operating PCR-SSP method to detect HLA-B27 is the true result, low cost, and the samples can be kePt for a long time. All that will be resulted in a quality evaluation in lab. At the same time, the method of flow cytometer provides us with facility, high Automation and objective result.
