海洛因对大鼠肾脏功能及核苷酸与氨基酸代谢相关酶活性的影响 被引量:7

摘要 目的研究海洛因对大鼠肾脏功能及代谢的影响。方法50只成年♀性Wistar大鼠平均分为5组,对照组腹腔注射生理盐水12d,两给药组分别腹腔注射海洛因3d和9d,两停药组腹腔注射海洛因9d后分别停药3d和9d。自动生化分析仪分析血浆尿酸(UA)、尿素氮(UN)、肌酐(CRE)及肾脏组织谷氨酸脱氢酶(GDH)含量,比色法检测肾脏组织腺苷脱氨酶(ADA)、黄嘌呤氧化酶(XOD)、谷氨酰胺合成酶(GS)及谷氨酸(Glu)含量。结果与对照组相比,两海洛因给药组血浆中CRE及UN均无明显升高,两停药组CRE升高(P<0.05),UN在停药9d后明显升高(P<0.01);两给药组大鼠血浆UA比对照组明显升高,两停药组大鼠血浆UA与对照组无差异;各实验组大鼠肾脏ADA含量均低于对照组;与对照组比,肾脏GS含量从给药3d开始升高(P<0.01),停药9d后恢复到对照组水平;肾脏GDH给药9d后明显低于对照组(P<0.01),停药9d后高于对照组(P<0.05);实验中XOD、Glu含量无变化。结论长期应用海洛因影响肾脏组织的代谢并损害肾脏功能。 Aim To study effects of heroin on kidney function and metabolism. Methods 50 adult female Wistar rats were divided randomly into 5 groups. The control group was intraperitoneal injected (ip) saline for 12 days. Two administration groups were ip heroin for 3 days and 9 days separately. Two withdrawal groups were ip heroin for 9 days followed by withdraw for 3 days and 9 days individually. Biochemistry autoanalyzer was applied to detect contents of uric acid ( UA), urea nitrogen ( UN ) and creatinine ( CRE ) in plasma and glutamate dehydrogenase(GDH) in kidney tissue. Contents of adenosine deaminase ( ADA), xanthine oxidase ( XOD ), glutamine synthetase ( GS ) and glutamic acid (Glu) were measured by colorimetric method. Results Plasma CRE in two withdrawal groups were both higher than that in control group (P 〈 0. 05). Plasma UN in 9 days withdrawal group was markedly higher than that in control group(P 〈0. 01 ). Plasma UA in two administration groups were increased compared with control group and no difference was found between two withdrawal groups and control group. ADA in kidney of four experimental groups were all increased significantly compared with control group. GS in kidney started to increase after 3 days heroin treatment ( P 〈 0. 01 ) and became equal to control group after 9 days withdrawal. GDH in kidney declined after 9 days heroin administration ( P 〈 0. 01 ) and increased after 9 days withdrawal( P 〈 0. 05 ) compared with control group. XOD and Glu content changed little during the experiment. Conclusion Long term heroin administration affects kidney metabolism and damage kidney function.
