摘要
植物细胞分离制样技术,原是应用于光学显微镜的细胞学的研究,我们将该技术中的固定、氧化、水解和分离程序与扫描电镜植物组织块常规制样方法中的脱水、临界点干燥程序相结合,形成了一种扫描电镜植物细胞分离制样方法,该方法的应用不仅使分离细胞的研究由光镜扩展至扫描电镜而且与扫描电镜植物组织块的常规方法等在细胞形态学研究方面可从互为对照和补充。
The sample preparation technique of plant cell separation used to be applied for cell obser-vation by light microscope. This paper described a method combined the procedures of fixation, oxidation, hydrolysis and seoaration in this technique with those of dehydration, critical point dryer etc. In the conventional method of plant tissue preparation to constitute a new method for SEM, The application of the new technique makes the research on separation of plant cell be developed from light microscopic level to SEM level. Therefore, it will be useful to enhance the research level in morphology and structure of plant cells and, meanwhile, make a supple-ment to the conventional method of preparing sample for SEM.
