摘要
目的:建立用高效液相色谱法同时测定血清中4种苯二氮芯卓类药物氯硝西泮、艾司唑仑、三唑仑、地西泮的方法。方法:血清样品在碱性条件下用乙醚(重蒸)提取,50℃水浴蒸干,残渣用流动相溶解,进样分析。色谱条件为ODS C18柱(5μm,250mm×4.6mm),流动相为甲醇-水-乙腈(53∶42∶5),流速为0.8mL.min-1,检测波长为254nm,柱温为35℃。结果:全部样品洗脱时间均不超过16min,药物之间能完全分离。4种药物平均回收率均在97%以上,样品在各自拟定的质量浓度范围内呈良好的线性关系,最大日内差和日间差分别为4.04%和9.61%。结论:本法提取简单快速,数据可靠,干扰少,适用于苯二氮芯卓类药血药质量浓度检测和中毒患者的急诊诊断。
OBJECTIVE To establish a rapid and simple HPLC method for the simultaneous determination of 4 kinds of benodiazepines including clonazepam, estazolam,midazolam and diazepam. METHODS The serum samples were extracted with ethyl ether,and theresidue was reconstituted in methanol. The mabile phase consisted of methanol-water -acetonitrile (53:42: 5). A discovery C18 column (250 mm×4. 6 mm,5 μm) was used. The detection wavelength was 254 nm. the column temperature was 35 ℃. RESULTS All components were eluted in 16 minutes and separated from each other. The relative recoveries of 4 drugs were above 97%. All components showed good linear relationship in the specified range. The max variation within-day and between-day were 4. 04%, 9. 61%. CONCLUSION The method is simple, accurate with few interferences and it can be used in monitoring four benzodiazepines in patients and can give an early diagnosis to save the poisoned patient.
