摘要
[目的]建立一种快速、简便、灵敏的测定血清总蛋白的方法。[方法]在非离子型表面活性剂聚氧乙烯月桂醚(Brij-35)存在下,用水溶性试剂偶氮胂Ⅲ(ArsenazoⅢ)作显色剂直接分光光度法测定血清总蛋白。[结果]该方法测定蛋白质,显色络合物的最大吸收波长为610nm,线性范围在0~56.2mg/L,表观摩尔吸光系数为3.60×10~5L.mol^(-1).cm^(-1)。回收率为96.1%~103.6%,批内相对标准差和批间相对标准差(RSD)分别为0.8%与2.0%,与双缩脲法比较相关性良好,Y=0.9882X+1.1007,r=0.9898,P﹥0.05。59名健康成年人血清总蛋白含量为(62.9~80.8)g/L(±2s)。[结论]该法血清用量少,线性范围宽,具有操作快速、简便、结果灵敏可靠等优点,适用于血清总蛋白的手工测定和自动分析。
[Objective] To establish a simple, sensitive and stable method of determining of total protein in serum by spectrophotemetry. [ Methods] Arsenazo Ⅲ was used as a coloring reagent to determine protein by spectrophotometry in the presence of surfactants Brij-35. [Results] The maximum absorption of the compound was at 610 van. The linearity of the test was 0-56.2 mg/L. The molar absorption coefficient was 3.60×10^5L·mol^-1·cm^-1. The recoveries ranged from 96.1% to 103.6%. The within-run and between-run CVs were 0.8%-1.5%and 1.5%-2.0%.Comparing this method (Y) with Biuret method, we obtained the regression equation Y = 0.9882X+1.1007, r = 0.9898, P 〉 0.05, the reference interval (x^-±2s) was (62.9- 80.8) g/L (n = 59) . [Conclusion] This method is simple, sensitive, rapid and accurate, and it's suitable for the routine manual or automatic analysis of total protein in serum for clinical application.
