摘要
目的探索马钱子制备癫痫动物模型的可能性及方法。方法分别采用单次、多次腹腔注射的方法予实验组大鼠以不同剂量的马钱子碱,观察大鼠注射后的行为学变化;同时用免疫组化法检测点燃鼠皮质和海马MAP2的表达,并与空白对照组及假模型组比较。结果接受单次腹腔注射91~110mg/kg大鼠均有癫痫发作,剂量越大,出现癫痫发作的可能性越大,但死亡率极高;分次注射相同剂量的马钱子碱,死亡率下降。对点燃鼠进行的MAP2测定发现其表达比空白对照组及假模型组明显升高,此与在癫痫患者脑组织中研究的结果相似。结论利用马钱子碱致痫性质来制备癫痫模型以研究人类的癫痫发作是可行的。
Objective: To identify the possibility and method of brucine in the establishment of brucine induced epilepsy model in rats according the fact that brucine could induced seizers in human. Methods: Observed the rats' behavioral changes by single intraperitoneal injection of brucine and repeated intraperitoneal injection of brucine at different dosages. The cerebral cortex and hippocampus of kindled rats were studied by using immunohistochemistry for MAP2 expression. Control brain tisssues were used for comparison. Results: In the groups injected with 91 mg/ kg - 110 mg/kg once, the seizures cold be observed, when we increased the dosage of injection, we observed more seizures. But the mortality rate was high. In contrast, the ratio of death decreased in repeated intraperitoneal injection groups injected the same dosage. The immunohistochemistry data showed that the expression of MAP2 in kindled rats was significant higher than those in the control groups, and the result was simlar to the research in the brain tissues of epilepsy patients. Conclusion: The study provided a rat epilepsy model induced by brucine. This model might be used in further research in human epilepsy.
