摘要
目的研究苯对小鼠胚胎肢芽发育和分化的影响,探讨苯的发育毒性。方法应用体外培养方法,对12天孕龄小鼠胚胎前肢芽进行培养,观察不同浓度的苯对肢芽骨发育和分化的影响。结果培养基中苯浓度达到75μmol/L以上时,前肢芽各软骨的发育和分化受到抑制,并且随着苯浓度的增加,肢体中各软骨的发育分化越差,Neubert评分越少,呈剂量-效应关系。结论苯为小鼠胚胎发育毒性物质,可抑制小鼠胚胎肢芽发育和分化。
Objective To observe the influence of benzene on mouse limb bud development and differentiation and evaluate the toxicity effect of benzene for limb buds of embryo mouse. Methods An invitro method was used to cultivate limb bud at embryo mouse 12 - day age to observe the influence of benzene in different concentration on mouse limb bud development and differentiation. Results The development and differentiation of mouse forelimb buds were inhibited treated with benzene at 75μmol/L and the Neubert score was decreased, with the concentration increasing, the inhibitory effect was more conspicuous, and there was the dose - effect relationship. Conclusion Benzene could inhibit the development and differentiation of embryo mouse limb bud in vitro. It might be a potential developmental toxic substance in mice.
