补脾方药对脾虚大鼠壁细胞CaM及Ca^(2+)/CaM-PK II活性的影响 被引量:13

摘要 目的:观察大黄脾虚模型大鼠壁细胞内CaM及CaCa2+/CaM-PK II活性的变化以及补脾方药党参、白术、补中益气汤对其的作用。方法:EDTA-Pronase酶消化法及Perco ll密度梯度离心分离和纯化大鼠胃壁细胞,根据的CaM的环核苷酸磷酸二酯酶检测法(PDE酶法)检测大鼠壁细胞内CaM活性;根据CaCa2+/CaM-PK II活性与激活底物磷酸化量成线性关系,测定其活性。结果:大黄脾虚模型大鼠壁细胞内CaM及CaCa2+/CaM-PKII活性均明显升高,补脾方药对其均有明显下调作用。结论:脾虚大鼠壁细胞处于高应激状态,党参、白术、补中益气汤等补脾方药可因对细胞内受体后信号传导物质的影响而调节细胞功能。 To observe the activity changes of CaM and Ca^2+/CaMⅡ in parietal cells of Pi-deficiency rats and the influence of Pi-invigorating recipe and herbs on them. Methods: Pronase-EDTA method was used to isolate rat gastric parietal cells. And the parietal cell purification could be achieved by nonlinear density gradient centrifugation using percoll. The activity of CaM and Ca^2+/CaMⅡ in parietal cells were detected according to the methods of reference articles. Results : The activity of CaM and Ca^2+/CaMⅡ increased in parietal cells of the Pi-deficiency group, and decreased in the treatment group of Pi-invigorating recipe and herbs. Conclusion : The parietal ceils of Pi-deficiency rat were in a high stress state. Once stimulated by receptor, the activity of intracellular signal substances increase which induce the enhanced biological effects.
