摘要
目的:探讨通络冻干粉针中有效成分栀子苷和芍药苷的药代动力学。方法:Beagle犬以高、中、低三个剂量分别单剂量静注给药,采用HPLC法测定不同时间Beagle犬体内栀子苷和芍药苷的血药浓度,以药动学计算软件对血药浓度-时间数据进行拟合。结果:高、中、低三种剂量分别单剂量静注给药后栀子苷、芍药苷在Beagle犬体内过程均呈二室模型,栀子苷高、中、低剂量组消除半衰期分别为113.5±41.5min,120.1±39.4min,161.0±22.6min,芍药苷为106.2±46.4min,132.7±46.8min,152.0±78.0min。结论:所建立的方法准确、灵敏度高,专属性好,可用于栀子苷和芍药苷的体内血浆药物浓度的同时分析。
Objective: A HPLC method was established for determination of genioside and paeoniflorin in plasma after iv Tongluo injection to dogs. Methods : After iv beagle dogs with high dosage, middle dosage, low dosage singly, HPLC was used to determine genioside and paeoniflorin in dogs' plasma at different times, and a pharmacokinetic computable procedure was used to process concentratoion-time data. Results : Beagle dogs were given high dosage, middle dosage, low dasage singly, and an open two-compartment model bes described the concentration time profile for genioside and paeoniflorin. The halflives of geinoside after i.v. high dosage, middle dosage and low dosage singly were 113.5 ± 41.5min, 120.1 ± 39.4min, 161.0 ± 22.6min, the halflives of paeoniflorin were 106.2 ± 46.4min, 132.7 ± 46.7min, 152.0 ± 78.0min. Conclusion : The analytical method was shown to be an accurate and sensitive way in detecting the in vivo blood concentration of genioside and paeoniflorin in plasma at the same time.
