摘要
目的探讨胰岛素对拟老年痴呆(AD)大鼠学习记忆能力及脑内胶质纤维酸性蛋白(GFAP)的影响。方法大鼠海马背侧微量注射甲基软海绵酸(大田酸),胰岛素侧脑室注射;水迷宫实验检测大鼠学习记忆能力;免疫组化观察大鼠脑内GFAP的表达。结果与对照组比较,模型组大鼠学习记忆能力下降,GFAP表达增多;与模型组相比,胰岛素组大鼠学习记忆能力明显提高,GFAP表达减少。结论胰岛素提高拟AD大鼠的学习记忆能力可能与其促进中枢神经系统生长发育,抑制GSK-3β和OA对tau蛋白的磷酸化作用,以及改善星形胶质细胞的功能有关。
Objective To investigate the effect through which insulin affect the learning and memory capabilities and GFAP of the Alzheimer's disease-like rats. Methods Okadaic acid ( OA ) was injected into the CA1 region of the rat hippocampus and the rats were injected with insulin. The learning and memory abilities of rats were assessed through Morris Water Maze behavioral test. We observed the expression of GFAP by immunohistoehemistry. Results Compared with the control rats, the capabilities of learning and memory were weakened significantly. The expression of GFAP positive neurons in the CA1 region increased in the model rats. In comparison with the model rats, the capabilities of learning and memory increased significantily, the expression of GFAP positive neurons in the CA1 region decreased in the rats injected with insulin. Conclusion Insulin can inhibit the function of GSK-313 and OA,and improve the function of astroeytes of Alzbeimer's disease-like rats,so it can increase their capabilities of learning and memory.
