补肾调冲含药血清对离体卵巢颗粒细胞Bcl-2、P53蛋白表达的影响 被引量:5

摘要 [目的]探讨补肾调冲方对大鼠离体卵巢颗粒细胞原癌基因蛋白表达的影响。[方法]采用血清药理学的方法,以不同含药量的大鼠血清与体外培养的卵巢颗粒细胞共同孵育24 h,用免疫组化法检测Bcl-2、P53蛋白的表达。[结果]补肾调冲含药血清可增强Bcl-2蛋白的阳性表达。[结论]原癌基因作为卵巢内局部调节因子,参与卵巢颗粒细胞凋亡过程。补肾调冲含药血清可通过促进卵巢颗粒细胞内抑癌基因的蛋白表达,抑制颗粒细胞的凋亡。 [Objective] To study the effect of BuShenTiaoChong (Tonifying kidney and regulating CHONG meridian) prescription on the expression of Bcl-2 and P53 proto-oncogene protein in isolated rat ovary granular cells (GC) in vitro. [Methods] Using a method of medicinecontaining serum the cultured in vitro ovary GC was co-cultured with rat serum containing different concentration of medicine for 24 hours. The expression of Bcl-2 and P53 oncogene protein in rat ovary granular cells was examined by immunohistochemistry. [Results] The serum with medicine could increase the positive expression of Bcl-2 oncogene protein in GC. [Conclusion] As the intra-ovarian local regulators Oncogene has involved in the process of apopto- sis of GC. The BushenTiaochong containing serum may inhibit apoptosis of GC through promoting the protein expression of anti-oncogene in ovary granular cells.
