摘要
目的]检测膀胱移行细胞癌(BTCC)组织中HIF-1α的表达情况,初步探讨HIF-1α在BTCC中表达的临床意义。[方法]应用免疫组织化学及RT-PCR技术检测BTCC组织中HIF-1α的蛋白和mRNA表达,并分析其表达与肿瘤临床分期、病理分级及复发的关系。[结果]免疫组织化学染色显示:12例正常膀胱组织未发现HIF-1α阳性表达,57例BTCC组织中有30例HIF-1α表达阳性,阳性表达率占52.63%。其表达与肿瘤的病理分级无关,但与肿瘤的临床分期(Tis—T1期的表达率与T2~T4期相比,P=0.026)及复发明显相关。RT-PCR检测显示:BTCC组织中HIF-1αmRNA的阳性表达率显著高于正常膀胱组织,但BTCC组织中HIF-1αmRNA的表达与其蛋白水平表达无明显相关性。[结论]在BTCC中,HIF-1α蛋白的表达明显上调,HIF-1α在BTCC的侵袭等生物学过程中具有重要的作用,HIF-1α可能可以用于临床监控BTCC的进展。
[Objective]To investigate the clinicopathological significance of HIF-1α expression in bladder transitional cell carcinoma(BTCC). [Methods]HIF-1α protein and mRNA expressions were determined using immunohistochemistry and reverse-transcriptase polymerase chain reaction(RT-PCR) respectively. The relationship between HIF-1α expression and various clinicopathological parameters including pathological grade and clinical stage was analyzed. [Results]Immunohistochemically, over-expression of HIF-1α was frequently (30 of 57 cases, 52.63%) detected in BTCC tissues , whereas no HIF-1α expression in 12 normal bladder tissues. HIF-1α expression was significantly associated with invasive bladder cancer(Tis-T1 versus T2- T4, P = 0. 026), but not with increased pathological grade. With RT-PCR technology,the positive expression rate of HIF-1α mRNA in BTCC group was higher than that in the normal bladder tissues group (P = 0. 033). There was no correlation between the expression of HIF-1α protein and the HIF-1α mRNA expression. [Conclusion] HIF-1α protein is frequently expressed in BTCC, HIF-1α may play a critical role in the invasion of BTCC and can be used to estimate its progression.
