摘要
目的 探讨肝癌形成过程中转化生长因子(TGF)-β1和端粒酶表达及其早期诊断价值。方法 雄性SD大鼠以2-乙酰氨基芴(2-FAA)喂饲制备肝癌模型,在肝细胞癌变过程中,观察肝脏总RNA变化,以病理组织学分析肝细胞形态学变化,采用酶联免疫吸附法(ELISA)分析肝细胞TGF-β1,以聚合酶链反应-ELISA分析端粒酶的动态变化。结果病理学证实大鼠在喂饲2-FAA后,正常肝细胞在诱癌早期发生颗粒样变性,中期阶段肝组织中出现不典型增生,后期可见大量癌巢结节,均为高分化肝细胞癌。在正常肝细胞及血清中仅见较低水平的TGF-β1和端粒酶表达。诱癌后,随着肝细胞组织形态的变化,肝组织及血清中TGF-β1和端粒酶表达水平逐渐增强,癌变组和癌前病变组均显著高于正常对照组(P〈0.05),且肝组织中TGF-β1、端粒酶和肝脏总RNA的表达呈显著正相关(P〈0.05)。结论 TGF-β1和端粒酶呈过表达与肝细胞癌变发生发展密切相关,且两者均有助于肝癌的早期诊断。
Objective To investigate the expression and early diagnostic values of transforming growth factor-betal (TGF-β1) and telomerase in hepatocarcinogenesis. Methods Hepatoma models were induced with 2-fluorenylacetamide (2-FAA,0.05%) in male SD rats. Morphological changes were observed by pathological examination (HE staining). Dynamic alterations of TGF-β1 and telomerase in liver or serum at different stages of canceration were analyzed by ELISA or PCR-ELISA, respectively. Results The rat hepatocytes showed granule-like denaturation at the early stages, then atypical hyperplasia at middle stage, and finally progression to tubercles of cancerous nest with highly differentiated hepatocellular carcinomas ( HCC ) during development of rat HCC by Histological examination. The increase of TGF-β1 and telomerase expression in liver and serum were found from normal liver to precancer- ous and cancerous tissues during rat hepatoma development, comparied with lower expression in normal controls. The positive relationship was found between liver total RNA and telomerase or TGF-β1 ( P 〈 0.05 ) at cancerization of rat hepatocytes. Conclusion The overexpressions of hepatic TGF-β1 and telomerase are closely related with the development of HCC, and both could be useful markers for early diagnosis of HCC.
