摘要
目的探讨白藜芦醇(resveratrol,Res)体外对胰腺癌MIAPaCa-2细胞增殖和细胞周期分布的影响。方法采用四甲基氨唑蓝法(MTT)检测Res处理后MIAPaCa-2细胞的增殖情况,用流式细胞仪分析经25,50,100,200μmol/L Res分别作用48 h后MIAPaCa-2细胞周期分布的改变。结果Res体外能显著抑制MIAPaCa-2细胞的生长增殖,且在一定范围内呈浓度依赖性。Res作用后细胞G0/G1期比例下降,S期比例上调,G2/M期未见规律性改变。结论Res在体外能显著抑制MIAPaCa-2细胞的增殖,引起细胞周期重新分布可能是Res抑制MIAPaCa-2细胞增殖的机制之一。
Objective It is to approach the influence of resveratrol on the proliferation and cell cycle distribution of MIA- PaCa- 2 cells in vitro. Methods Methyl thiazolyl tetrazolium method (MTT) was used to detect the cell growth status of MIAPaCa - 2 cells after treatment by resveratrol. Cell cycle distributions of MIAPaCa - 2 cells were analyzed by flow cytometry (FCM) after cells were treated with 25μmol/L, 50μmol/L, 100 μmol/L and 200μmol/L resveratrol for 48h respectively. Results Resveratrol inhibited the proliferation of MIAPaCa -- 2 cells in a concentration-dependent manner as measured by MTT method. Resveratrol decreased the percentage of cells in the G1 phase with a concomitant increased in the proportion of cells in the S phase. Conclusion These findings suggest that resveratrol may have a potent anti-proliferative effect on MIAPaCa - 2 cells, the mechanisms of which may be related to the induction of re-distribution of cell cycle.
