摘要
应用特异性的a-rLHβ抗血清,以ABC技术显示垂体黄体生成素(LH)细胞,通过计算机图像处理系统对LH细胞的面积、细胞内液泡面积和细胞形状等参数进行定量和统计分析,获得了大鼠正常动情周期各时期典型的LH细胞学图像。结果表明:LH的早期贮存和基础分泌主要同细胞内颗粒变化有关,而LH的大量贮存和大量释放同细胞内液泡的变化以及由此产生的细胞大小、细胞形状和细胞结构等形态变化密切相关。
n this study, immunostaining was performed with a specific a r LH antibody by ABC method in the rat′s pituitary at the five different phases during the estrus cycle. The average area of LH cells and vacuoles, and the shape of the cells were calculated by medical image processing system I. Final data were analyzed with SAS statistical software by VAX II computer. The results were as follows: (1) In diestrus and the early stage of proestrus, a huge amount of LH were stored in LH cells, and about 49.01% and 43.9% of the cells contained large vacuoles respectively. The area of LH cells increased with enlargement of the vacuolar size, and was significantly larger than those in other phases of the estrus cycle. In the meanwhile, most of LH cells were approximate to the round in shape; (2) In the late stage of proestrus, with the LH release peak, LH cells containing large vacuoles decreased to 10.98%. With the significant decrement of the vacuoles, an important change of LH cells in shape was characterized by appearing a large amount of irregular and smaller cells; (3) After LH peak release, from estrus to metestrus, synthesis and storage of LH were restarted. LH cells with medium size were predominant, and contained small vacuoles and vesicles. With the increment of LH secretory granules, most of LH cells gradually approximated to circle, and the number of irregular cells was decreased. In conclusion, the storage of LH relating to changes of granules was found in early stage of storage (estrus and metestrus). The storage and the release of a huge amount of LH in diestrus and proestrus closely corrected with the structural changes of the vacuoles.
