摘要
用自制的溴代丙酮与苯亚磺酸钠反应合成1-苯砜基-2-丙酮,然后利用面包酵母在有机溶剂中不对称催化还原1-苯砜基-2-丙酮,制备了光学活性的手性化合物(S)-1-苯砜基-2-丙醇,此反应具有操作简便、光学产率高等优点,其化学产率可达94%以上,对映体过量值为100%。
In this paper reaction of asymmetric reduction of 1 - Phenylsulfonyl - 2 - Propanone by baker's yeast in organic solvents has been studied. First, three levels and four factors orthogonal experimental were applied to probe into the condition of this reaction. And then, effects of water quantity, substrate concentration, pH value and reaction time on the reaction yield were systemically studied ulteriorly. At the same time, the kinetics of the reaction was also explored. The results showed that the yield was up to as high as 94% under the optimum conditions, and the enantiomefic excess approached 100%. Data of kinetics were accorded with Michaelis - Menton enzyme catalyze mechanism. The maximum reaction ate and constant of Michaelis - Menton were obtained.
