摘要
目的观察玉红膏对表皮干细胞增殖、分化的影响,探讨玉红膏促进创面愈合的作用机理。方法采用大鼠全层皮肤缺损模型,完全随机法分成玉红膏组、京万红组、模型组、正常对照组。于造模后第1、3、5、7、14、21天6个时间点观察记录创面的愈合情况、完全愈合时间并取材。行表皮干细胞标志物整合素β1、转录因子p63的免疫组化染色,比较各组各观测点OD值及阳性细胞数的差异。结果玉红膏组平均愈合时间[(13.5±0.9)d]与京万红组[(12.6±0.9)d]比较无明显差别,均优于模型组[(19.1±2.6)d],P<0.05;大体观察可见玉红膏组早期创面渗出物较多且浓稠;整合素β1、转录因子p63的免疫组化染色结果显示,玉红膏组、京万红组、模型组在造模后整合素β1OD值、转录因子p63表皮层阳性细胞数均有升高,高于正常组,且差异有意义(P<0.05);玉红膏组、京万红组在第7天达到峰值,峰值出现的时间早于模型组,峰值的强度高于模型组,差异有意义(P<0.05);玉红膏组这2个指标前14d均高于模型组(P<0.05);京万红组前7d高于模型组(P<0.05),第14天与模型组无差异,至创面完全愈合的第21天3组无差别。结论玉红膏有促进创面愈合的作用,机理可能与其诱导创缘残留的表皮干细胞增殖分化有关。
Objective To observe the effect of Yuhonggao on proliferation and differentiation of epidermal stem cells and investigate the mechanism of promoting wound healing. Methods The whole layer skin defection rat was used and randomly divided into 3 groups. Yuhonggao group, Jingwanhong group, model group, with normal rats as control, The condition of skin wound surface and healing time were observed and recorded, and then draw the wound skin. The p63 and β1 integrin expression, the marker of epidermal stem cells, were measured and compared by immunohistochemistry staining. Average wound healing time hadn't significant difference between the Yuhonggao group [(13.5±0.9)d] and Jingwanhong group [(12.6±0.9)d], which were better than model group (P〈0.05). By gross observing, there were more and thick secretion of Yuhonggao group in initial stage. The results of immunohistochemistry staining of p63 and β1 integrin illustrate the measured value of optical density of β1 integrin and positive cell population in epidermis layer of p63 advanced after damaged in Yuhonggao group, Jingwanhong group and model group, which were higher than normal group (P〈0.05). The max showed up after 7 days in Yuhonggao group and Jingwanhong group, it's not only earlier than model group but also harder than it (P〈0.05). After damaged 14 days, the 2 indexes of Yuhonggao group were higher than model group. It's 7 days in Jingwanhong group (P〈0.05), it hadn't significant difference in the 14th day, there was no difference among 3 groups on 21 th day. Conclusion Yuhonggao can accelerate skin wound healing and possibly relate with promoting proliferation and differentiation of epidermal stem cells in edge of skin wound surface.
