吡格列酮对非肥胖性糖尿病小鼠视网膜细胞凋亡的影响 被引量:2

摘要 目的观察吡格列酮对早期糖尿病小鼠视网膜细胞凋亡的影响。方法非肥胖性糖尿病(NOD)雌性小鼠20只随机分为对照组(n=10)和吡格列酮干预组(n=10)。对照组给予普通饲料,干预组用加入0.02%吡格列酮的饲料喂养。2组分别在4周时和12周处死小鼠,摘除眼球并制作组织切片,用TUNEL法检测视网膜细胞的凋亡。电镜观察视网膜超微结构改变。结果干预组小鼠视网膜神经节细胞和内皮细胞凋亡数目明显较对照组减少(P<0.01)。结论吡格列酮可抑制NOD雌鼠视网膜细胞凋亡,对视网膜的神经元病变和微血管病变有治疗作用。 Objective To observe the effect of ploglitazone on retinal cells apoptosis in early diabetic mice. Methods Non-obese diabetic (NOD) mice were randomly divided into control group ( n = 10) and pioglitazone intervention group ( n = 10) .The mice in control group were fed with common food, however, the mice in pioglitazone intervention group were fed with food in which 0.02% pioglitazone were added.The mice in both groups were killed at 4 weeks and 12 weeks, and eyeballs of mice were taken out for detecting retinal cell apoptosis by TUNEL method, and the changes of retinal uhrastructure were observed by electronic microscope. Results The retinal ganglion cells and endothelium apoptosis decreased significantly in pioglitazone intervention group, as compared with that in control group ( P 〈 0.01 ). Conclusion Pioglitazone can inhibit retinal cell apoptosis of diabetic mice and have therapeutic effect on pathological changes of neuron of retina and microangiopathic changes.
